In August 2026, “a systematic review and pooled analysis of phase 3 efficacy and integrated safety across clinical trials and real-world evidence” was published by the British Society of Haematology.
It found that post-ruxolitinib JAK inhibitors demonstrate distinct efficacy and safety profiles, and treatment selection should be guided by a patient’s dominant clinical phenotype.
In Australia, please note that whilst ruxolitinib, momelotinib and fedratinib are available and on the PBS, Pacritinib is not available.
The authors conclude that “Post-ruxolitinib JAKi selection is best guided by the dominant clinical phenotype rather than a single ranked drug list. Looking ahead, biomarker-driven patient selection, combination strategies incorporating anti-fibrotic or erythropoiesis-supporting agents, and prospective registries with harmonized end-points are the three priorities most likely to refine current practice.”
